FCS Express 7 allows the user to validate the quality of the actual spectral signatures of single stained controls against the expected spectral signatures with the FCS Express-EasyPanel integration. This feature is available to all FCS Express 7 users.

 

Users would first need to create an Unmixing Matrix in FCS Express 7.

 

The Validate with EasyPanel button is located in the Unmixing Matrix tab of the Compensation and Unmixing dialog (Figure 9.58, rightmost button in the red box below).

This button will open a new tab in the EasyPanel website that shows the Actual vs Expected Similarity of each single stained controls. An example of which is depicted in Figure 9.59.

EasyPanel has the expected flurochrome spectral signature on your flow cytometer and will plot your actual spectral signature against this data. Here you can then see the similarity score of each fluorochrome in your Unmixing.

 
Actual vs Expected Similarity Score

Score = 1: Actual spectral signature is the same as the Expected spectral signature.

Score < 1: Indicates a difference between the Actual spectral signature and the Expected spectral signature.

Some causes of a low similarity score are contamination of the single stained controls, wrong single stained controls, or dye/fluorophore degradation due to poor storage or exposure to light.

 

EasyPanel account

While you can use the FCS Express-EasyPanel integration without an EasyPanel account, if you are logged into an EasyPanel account when validating your spectral signatures, you will be able to review your single stained controls over time from previous exports and take your Autofluoresence Signature(s) forward to future Panel Design or Panel Reviews.
